目的:研究中药何首乌主要有效成分二苯乙烯苷(THSG)对多柔比星(DOX)损伤心肌细胞的保护作用及机制。方法:分离培养乳鼠心肌细胞,观察DOX单独及与THSG联合处理对细胞凋亡、活性氧自由基(ROS)生成及线粒体膜电位的影响。结果:THSG能够剂量依赖性地减少DOX诱导的心肌细胞凋亡、ROS生成及线粒体膜电位的下降。结论:THSG对DOX诱导的心肌细胞损伤具有保护作用,且作用与其能够减少ROS生成、抑制线粒体损伤介导的凋亡有关。
Objective: To study the protective effect of stilbene glycoside (THSG) on cardiomyocytes injured by doxorubicine (DOX) and its mechanism. Methods: The neonatal rat cardiomyocytes were isolated and cultured. The effects of DOX alone and combined with THSG on apoptosis, reactive oxygen species (ROS) production and mitochondrial membrane potential were observed. RESULTS: THSG reduced DOX-induced cardiomyocyte apoptosis, ROS production and mitochondrial membrane potential in a dose-dependent manner. CONCLUSION: THSG has a protective effect on DOX-induced cardiomyocyte injury and its role is related to its ability to reduce ROS production and inhibit mitochondrial damage-mediated apoptosis.
