目的研究自体骨髓间充质干细胞(bone marrow stem cells,BMSCs)复合富血小板纤维蛋白(platelet-rich fibrin,PRF)对兔拔牙窝骨缺损新骨形成的促进作用。方法将2月龄雄性新西兰兔27只,随机分3组,每组9只,建立动物模型,均在全身麻醉下微创拔除下颌左侧中切牙。A、B和C组分别为BMSCs复合PRF、单一PRF和空白对照。每组3只于3个时间点,即第4、8、12周,在骨缺损部位取材,拍摄X线片观察骨缺损区骨质的愈合及改建情况。同时制作HE染色的组织切片,进行骨计量观察。结果观察和使用灰度值统计软件进行分析的结果表明,在4、8、12周A组缺损区骨密度明显高于B、C组(P<0.01)。A组与B、C组的骨计量学比较分析显示骨小梁宽度、缺损区骨量、成骨细胞数差异均有统计学意义(P<0.01)。结论自体BMSCs复合PRF应用于兔牙槽骨缺损区能有效促进新骨形成。
OBJECTIVE: To investigate the effect of platelet-rich fibrin (PRF) combined with autologous bone marrow stem cells (BMSCs) on the formation of new bone in rabbit’s socket defect. Methods 27 male New Zealand 2-month-old New Zealand rabbits were randomly divided into 3 groups (9 rats in each group). Animal models were established by minimally invasive extraction of mandibular central incisor under general anesthesia. Groups A, B and C were BMSCs combined PRF, single PRF and blank control, respectively. Three rabbits in each group were taken at 3 time points, ie the 4th, 8th and 12th weeks, at the bone defect site. X-ray films were taken to observe the healing and remodeling of the bone in the bone defect area. At the same time making HE-stained tissue sections for bone metrology observation. The results of observation and gray value statistical software analysis showed that the bone mineral density in group A at 4, 8 and 12 weeks was significantly higher than that in group B and C (P <0.01). A comparative analysis of bone metrology between groups A and B and C showed that the width of trabecular bone, the amount of bone defect in the defect area and the number of osteoblasts were all statistically significant (P <0.01). Conclusions Autologous BMSCs combined with PRF can effectively promote new bone formation in rabbit alveolar defects.
