目的 探讨区域性动脉灌注化疗对胰腺癌bcl 2、bax基因表达的影响 ,寻找胰腺癌治疗的新手段。方法 应用TUNEL法检测 3 2例未经区域性动脉灌注化疗和 19例经区域性动脉灌注化疗的胰腺癌细胞凋亡指数 (AI) ;并采用免疫组织化学方法检测区域性动脉灌注化疗对胰腺癌细胞bcl、bax基因表达的影响。结果 区域性动脉灌注化疗组胰腺癌细胞AI比未区域性动脉灌注化疗组明显升高 ( 0 65 %vs 1 10 % ,P <0 0 5 ) ;与未区域性动脉灌注化疗组相比 ,区域性动脉灌注化疗组肿瘤细胞bcl 2基因表达明显减少 ( 3 6 7%vs 75 0 % ,P <0 0 5 ) ,而bax基因的表达显著增加 ( 73 6%vs3 4 0 % ,P <0 0 5 )。结论 区域性动脉灌注化疗可以诱导胰腺癌肿瘤细胞凋亡的增加 ,是胰腺癌综合治疗的有效措施之一。
Objective To investigate the effect of regional arterial infusion chemotherapy on the expression of bcl 2 and bax gene in pancreatic cancer and to find a new method for the treatment of pancreatic cancer. Methods TUNEL method was used to detect the apoptotic index (AI) of 32 pancreatic cancer cells without regional arterial infusion chemotherapy and 19 with regional arterial infusion chemotherapy. Immunohistochemistry was used to detect the effect of regional arterial infusion chemotherapy on pancreatic cancer Effects of bcl and bax gene expression in cells. Results The AI of pancreatic cancer cells in the regional arterial infusion chemotherapy group was significantly higher than that in the non-regional arterial infusion chemotherapy group (0 65% vs 1 10%, P 0 05). Compared with the non-regional arterial infusion chemotherapy group, The expression of bcl-2 gene was significantly decreased in tumor-infused chemotherapy group (36.7% vs 75.0%, P <0.05), while the expression of bax gene was significantly increased (73.6% vs. 340.0%, P <0 0 5). Conclusion Regional arterial infusion chemotherapy can induce apoptosis of pancreatic cancer cells and is one of the effective measures for the comprehensive treatment of pancreatic cancer.
