目的:报道了人体内艾司唑仑及其主要代谢物4′羟基艾司唑仑的电喷雾离子阱质谱鉴定法。方法:采用固相萃取或液-液萃取从生物基质中提取待测物,然后将其直接导入ESI离子源进行质谱分析。结果:选择m/z295和m/z311为母离子,分别进行MS2和MS3级质谱分析,以它们生成的子离子(MS2:m/z267,MS3:m/z240和205)及其主要代谢物(MS2:m/z283,MS3:m/z256和221)来鉴定母体药物。结论:本法专属性强,灵敏度高(母体药物的LOD为05ng·mL-1),可作为法医毒物分析的一种方法。
OBJECTIVE: The electrospray ion trap mass spectrometry method for the determination of estazolam and its major metabolite 4’-hydroxy-estazolam in human is reported. Methods: The analytes were extracted from the biological matrix by solid-phase extraction or liquid-liquid extraction and then directly introduced into the ESI ion source for mass spectrometry analysis. Results: Mass spectra of m / z295 and m / z311 were selected as MS1 and MS3, respectively. Based on the mass spectra of their daughter ions (MS2: m / z267, MS3: m / z240 and 205) and their major metabolites MS2: m / z 283, MS3: m / z 256 and 221) to identify the parent drug. Conclusion: This method is highly specific and sensitive (LOD of parent drug is 0.5 ng · mL-1), which can be used as a method of forensic toxicological analysis.
