目的:将水性二相系统通过高速逆流色谱进行牛膝高分子物质中多糖成分的纯化研究。方法:以聚乙二醇与磷酸钾缓冲溶液组成水性二相系统,作为高速逆流色谱的固定相和流动相,对牛膝的高分子物质进行分离纯化。结果:125%聚乙二醇(PEG)1000与16%磷酸钾缓冲溶液(pH68)组成的水性二相系统,在XaxisCPC高速逆流色谱仪上,可以成功地分离出多糖部分和蛋白多糖部分。结论:借助高速逆流色谱,水性二相系统可以进行中药牛膝多糖的分离和纯化。
OBJECTIVE: To purify polysaccharides from Achyranthes bidentata polysaccharides by high performance countercurrent chromatography in an aqueous two-phase system. Methods: An aqueous two-phase system consisting of polyethylene glycol and potassium phosphate buffer was used as stationary phase and mobile phase in high-speed countercurrent chromatography to separate and purify achyranthes bidentata macromolecules. Results: The aqueous two-phase system composed of 12.5% polyethylene glycol (PEG) 1000 and 16% potassium phosphate buffer solution (pH6.8) could successfully separate the polysaccharide fraction on the XaxisCPC high-speed counter current chromatograph And proteoglycan parts. Conclusion: With high-speed countercurrent chromatography, aqueous two-phase system can be used for the separation and purification of Achyranthes bidentata polysaccharides.
