草鱼出血病是我国淡水渔业的一大病害,我所与长江水产所协作开展对草鱼出血病病毒的分离,防治和分子生物学的研究。本文侧重介绍利用电子显微镜对草鱼出血病病毒的形态和超微结构的研究。材料与方法病毒制备成悬液,滴于火棉胶炭膜铜网上待干后,2%磷钨酸负染。取病鱼的肝、脾、肾组织和病变细胞,用2%戍二醛和1%锇酸双固定,苯二甲酸二丙烯酯包埋,LKB—8800型超薄切片,于JEM—100C电子显微镜观察。
Grass carp hemorrhagic disease is one of the major diseases of freshwater fishery in China. In collaboration with the Yangtze River Fisheries Institute, we conducted the research on the isolation, prevention and molecular biology of grass carp hemorrhagic disease virus. This article focuses on the use of electron microscopy of grass carp hemorrhagic disease virus morphology and ultrastructure. Materials and Methods The virus was prepared into a suspension, which was dropped onto a sponge carbon film copper net to be dried, and then 2% phosphotungstic acid was negatively stained. The liver, spleen, kidney and diseased cells of diseased fish were fixed with 2% glutaraldehyde and 1% osmium tetroxide, embedded in dicaprylyl phthalate, ultrathin sections of LKB-8800, Microscopic observation.
