目的:观察丝裂霉素(MMC)联合雷帕霉素(RAPA)对体外培养的人膀胱癌T24细胞抑制作用是否比单药大。方法:体外培养人膀胱癌T24细胞,通过MTT实验及相关公式计算,得出MMC和RAPA的IC50值,以此值及相关药量应用于膀胱癌T24细胞,经24、48h,比较两种药物单用及联合用药(全量及半量)对癌细胞的抑制率,得出抑制率最高的药物(两种单药或联合用药)。采用方差分析、SNK-q检验分析得出结果。结果:MMC、RAPA的IC50值分别为12μg/ml、125μg/L(最后实验所用)。两种药物单用及联合用药的24h抑制率最高的是MMC联合RAPA,全量及半量联合的抑制率差异无统计学意义,两者抑制率分别为63.64%及73.68%;MMC联合RAPA比单药的抑制率高,差异有统计学意义;与单药48h抑制率比较,MMC联合RAPA的抑制率差异无统计学意义(其中MMC联合RAPA全量联合的抑制率最大,为98.53%),但是高于RAPA的抑制率,差异有统计学意义。结论:MMC联合RAPA对体外培养的人膀胱癌T24细胞抑制作用比单药大。
Objective: To observe whether mitomycin (MMC) and rapamycin (RAPA) inhibit the proliferation of human bladder cancer T24 cells in vitro than single drug. Methods: Human bladder cancer T24 cells were cultured in vitro. IC50 values of MMC and RAPA were obtained by MTT assay and correlation formula. T24 cells were used in this study. Twenty-four and 48 hours after the two drugs were compared Single and combined medication (full and half) of the inhibition of cancer cells, resulting in the highest inhibition rate of the drug (two single or combination). ANOVA, SNK-q test results. Results: The IC50 values of MMC and RAPA were 12μg / ml and 125μg / L, respectively (for the last experiment). The combination of MMC and RAPA showed the highest inhibitory rate of 24h combined with MMC and the combination of MMC and RAPA. There was no significant difference in the inhibition rates between the two groups (63.64% and 73.68%, respectively) (P <0.05). Compared with the inhibition rate of single drug 48h, there was no significant difference in the inhibition rate of MMC combined with RAPA (the largest inhibition rate of MMC combined with RAPA was 98.53%), but higher than RAPA inhibition rate, the difference was statistically significant. CONCLUSION: MMC combined with RAPA has a greater inhibitory effect on human bladder cancer T24 cells than monotherapy.
