目的:观察中药乳康饮对移植瘤小鼠乳腺癌血管内皮生长因子C/D(VEGF-C/D)、血管内皮生长因子受体(VEGFR-3)mRNA表达的影响,探讨其抑制乳腺癌转移的机制。方法:30只BALB/c裸鼠乳腺原位种植人乳腺癌细胞株MDA-MB-435S,建立乳腺癌自发性转移模型,随机分为模型对照组,5-FU对照组,乳康饮小、中、大剂量组,乳康饮+5-FU组,每组5只。模型对照组给予0.9%氯化钠溶液0.4mL/d灌胃;5-FU对照组给予5-FU 30mg·kg-1·d-1腹腔注射;乳康饮小、中、大剂量组分别给予乳康饮18、45、90g·kg-1·d-1灌胃;乳康饮+5-FU组给予5-FU注射液30mg·kg-1·d-1腹腔注射,同时给予乳康饮45g·kg-1·d-1灌胃,均每天1次,连续6周。用药后检测肿瘤质量和腋窝淋巴结转移抑制率,肿瘤组织VEGF-C/D、VEGFR-3基因的表达。结果:乳康饮各剂量组及乳康饮+5-FU组肿瘤质量与模型对照组比较明显减小(P<0.01),乳康饮+5-FU组抑瘤率及腋窝淋巴结转移抑制率明显高于其他药物组(P<0.05)。乳康饮各剂量组VEGF-D mRNA表达显著下调,与模型对照组比较,差异均有统计学意义(P<0.05,P<0.01)。乳康饮大剂量组和乳康饮+5-FU组VEGF-C mRNA、VEGFR-3 mRNA表达量下调,与模型组比较,差异有统计学意义(P<0.05,P<0.01)。结论:乳康饮可能通过干预VEGF-C/D、VEGFR-3 mRNA的表达,影响淋巴管的生成,抑制乳腺癌淋巴转移。
Objective: To observe the effect of Rukangyin on the expression of VEGF-C / D and VEGFR-3 mRNA in breast cancer in mice and to investigate its inhibitory effect on breast cancer Transfer mechanism. After treatment, the detection of tumor mass and axillary lymph node metastasis inhibition rate, tumor tissue VEGF-C / D, VEGFR-3 gene expression. Results: The tumor weight of Rukangyin + 5-FU group was significantly lower than that of the model control group (P <0.01), the inhibition rate of tumor and the rate of axillary lymph node metastasis of Rukangyin + 5-FU group Significantly higher than other drugs group (P <0.05). The expression of VEGF-D mRNA was significantly down-regulated in each dose of Rukangyin compared with the model control group (P <0.05, P <0.01). The expression of VEGF-C mRNA and VEGFR-3 mRNA in high-dose Rukangyin group and Rukangyin + 5-FU group were significantly lower than those in model group (P <0.05, P <0.01). Conclusion: Rukang Yin may interfere with the expression of VEGF-C / D and VEGFR-3 mRNA, affect lymphangiogenesis and inhibit lymphatic metastasis of breast cancer.
