目的 观察N-糖链抑制剂衣霉素(TM)对肾小球系膜细胞(GMC)增殖、凋亡及纤连蛋白(FN)的影响。方法 用流式细胞仪和透射电镜观察 GMC凋亡。四唑氮盐法(MTT法)测定TM对GMC增殖的影响。间接竞争抑制酶联免疫吸附法观察TM对细胞外基质(EMC)成分FN的影响。结果TM可抑制GMC的增殖(与对照组相比P<0.05),且呈剂量和时间依赖性;并可诱导GMC凋亡,除0.1μg/ml组外(P>0.05),其它处理组都可增加细胞凋亡率,且随时间延长和剂量增加呈上升趋势(P<0.05),电镜下可见GMC呈典型的凋亡形态学改变。与对照组比较,TM可减少上清FN含量(P<0.05)。结论 TM能够抑制GMC增殖,诱导细胞凋亡且呈剂量和时间依赖性,并可减少细胞外基质(ECM)生成。
Objective To investigate the effects of tunicamycin (TM), an inhibitor of N-glycans, on proliferation, apoptosis and fibronectin (FN) in glomerular mesangial cells (GMCs). Methods GMC apoptosis was observed by flow cytometry and transmission electron microscopy. Effect of TM on GMC proliferation by MTT method. Indirect competitive inhibition enzyme-linked immunosorbent assay was used to observe the effects of TM on FN, an extracellular matrix (EMC) component. Results TM could inhibit the proliferation of GMC (P <0.05 compared with the control group) in a dose- and time-dependent manner. The apoptosis of GMCs was also induced by TM, except for 0.1μg / ml group (P> 0.05) (P <0.05). The morphological changes of apoptotic cells were observed under electron microscope. Compared with the control group, TM can reduce the FN content of the supernatant (P <0.05). Conclusion TM can inhibit GMC proliferation, induce apoptosis in a dose and time-dependent manner, and can reduce the extracellular matrix (ECM) production.
