目的建立HPLC同时测定黄石感冒片中阿魏酸、芦丁、大黄酸、大黄素和大黄酚的含量。方法采用HPLC,色谱柱为Welch Topsil-C18柱(4.6 mm×250 mm,4μm);以甲醇-0.1%磷酸溶液(78∶22)为流动相;流速:1.0 m L·min?1;柱温:30℃;检测波长:280 nm。结果阿魏酸、芦丁、大黄酸、大黄素和大黄酚峰线性范围分别为0.020 82~0.416 3μg·m L?1(r=0.999 7),0.011 32~0.278 3μg·m L?1(r=0.999 3),0.017 22~0.344 3μg·m L?1(r=0.999 5),0.015 79~0.315 8μg·m L?1(r=0.999 6)和0.051 34~1.027μg·m L?1(r=0.999 9),平均加样回收率分别为97.4%(RSD=1.1%),95.0%(RSD=0.88%),97.5%(RSD=1.3%),97.4%(RSD=1.4%)和96.3%(RSD=0.87%)。结论新建方法简便、准确,可用于黄石感冒片的定量分析方法。
Objective To establish an HPLC method for the simultaneous determination of ferulic acid, rutin, rhein, emodin and chrysophanol in Huangshi. Methods The column was Welch Topsil-C18 column (4.6 mm × 250 mm, 4 μm) with methanol-0.1% phosphoric acid solution (78:22) as mobile phase at a flow rate of 1.0 mL · min -1. The column temperature : 30 ℃; detection wavelength: 280 nm. Results The linear ranges of ferulic acid, rutin, rhein, emodin and chrysophanol were 0.020 82 ~ 0.416 3 μg · m L -1 (r = 0.999 7), 0.011 32 ~ 0.278 3 μg · m L -1 (r = 0.999 3), 0.01722 ~ 0.344 3μg · m L -1 (r = 0.999 5), 0.01579 ~ 0.315 8μg · m L -1 (r = 0.999 6) and 0.051 34 ~ 1.027μg · m L -1 r = 0.999 9). The average recoveries were 97.4% (RSD = 1.1%), 95.0% (RSD = 0.88%), 97.5% (RSD = 1.3%), 97.4% % (RSD = 0.87%). Conclusion The new method is simple and accurate and can be used for the quantitative analysis of cold tablets in Huangshi.
