目的研究河南地区阴道毛滴虫临床分离株与人型支原体共生情况,并观察其共生对阴道毛滴虫AP33基因序列的影响。方法应用人型支原体及AP33基因的特异性引物对41株阴道毛滴虫临床分离株进行PCR扩增,扩增产物进行1.5%琼脂糖凝胶电泳检测,对AP33基因扩增产物测序,测序结果与GenBank已知序列比对,绘制进化树。结果有34株阴道毛滴虫扩增出人型支原体的特异性片段,大小为334bp;所有虫株均扩增出AP33基因特异性片段,碱基序列相似性为96.8%~99.3%,有多个位点发生突变。进化树分析提示人型支原体的共生对AP33基因序列有一定影响。结论河南地区阴道毛滴虫临床分离株与人型支原体共生具有普遍性,其共生对阴道毛滴虫的基因序列有一定影响。
Objective To study the symbiosis between Trichomonas vaginalis clinical isolates and Mycoplasma hominis in Henan Province and to observe their symbiosis with Trichomonas vaginalis AP33 gene sequence. Methods 41 strains of clinical isolates of Trichomonas vaginalis were amplified by PCR using primers specific to Mycoplasma hominis and AP33. The amplified products were detected by 1.5% agarose gel electrophoresis. The amplified products of AP33 gene were sequenced and sequenced Phylogenetic tree was constructed by aligning with known sequences in GenBank. Results Thirty-four Trichomonas vaginalis strains amplified specific fragments of Mycoplasma hominis, with a size of 334bp. The AP33 gene-specific fragments were amplified from all the isolates, and their nucleotide sequence similarity was 96.8% -99.3% A mutation occurred. Phylogenetic tree analysis suggested that Mycoplasma hominis symbiosis had some influence on the AP33 gene sequence. Conclusions The clinical isolates of Trichomonas vaginalis in Henan Province are common with Mycoplasma hominis. Their symbiosis has some influence on the gene sequence of Trichomonas vaginalis.
