目的:构建壳聚糖-胶原-TGF-β微球复合材料,探讨其用于牙周组织再生的可行性。方法:水包水复乳溶媒萃取法制备TGF-β微球,冷冻干燥法制备复合支架,扫描电镜观察。细胞培养后测量相关收缩率;MTT检测细胞增殖;共聚焦显微镜观察细胞三维生长。结果:支架呈现多孔穿通结构。细胞培养后无显著体积收缩,MTT检测实验组细胞增殖能力显著,共聚焦电镜下细胞密度显著高于对照组。结论:壳聚糖-胶原负载TGF-β微球的复合支架促进牙周组织再生具有可行性。
OBJECTIVE: To construct chitosan-collagen-TGF-β microspheres composite and explore its feasibility for periodontal tissue regeneration. Methods: TGF-β microspheres were prepared by double emulsion in water-in-water and the composite scaffolds were prepared by freeze-drying method. The relative contractility was measured after cell culture; the proliferation of cells was detected by MTT; the three-dimensional growth of cells was observed by confocal microscopy. Results: The stent showed porous structure. No significant volumetric shrinkage was observed after culture. MTT assay showed that the cell proliferation ability of the experimental group was significant, and the cell density under confocal electron microscopy was significantly higher than that of the control group. Conclusion: The composite scaffold loaded with TGF-β microspheres loaded with chitosan and collagen can promote periodontal tissue regeneration.
