目的观察成纤维细胞生长因子受体3(fibroblast growth factor receptor 3,FGFR3)在坐骨神经分支选择结扎后神经病理性疼痛中大鼠脊髓的表达变化。方法 40只SD雄性大鼠,分为假手术对照组(sham)和手术组(SNI),每组20只。使用坐骨神经分支选择结扎方式建立大鼠神经病理性疼痛模型,观察sham组和SNI组大鼠术前1 d以及术后1、3、5、7 d的行为学改变并检测2组大鼠的机械痛阈,采用免疫荧光双标观察2组大鼠术后7 d脊髓背角星形胶质细胞FGFR3和胶质纤维酸性蛋白(glial fibrillary acidic protein,GFAP)的共表达,Western blot法检测SNI术后各时间点2组大鼠FGFR3蛋白表达。结果术前1 d 2组大鼠机械痛阈无明显差异(P>0.05),术后1、3、5、7 d SNI组大鼠机械痛阈值分别为(6.67±2.12)、(3.17±0.99)、(2.33±0.65)、(1.75±0.31),相应时间点sham组大鼠机械痛阈值分别为(12.75±2.83)、(12.33±2.84)、(12.08±2.57)、(11.50±2.65),SNI组大鼠机械痛阈在各时间点较sham组明显降低(P<0.05);术后7 d患侧脊髓背角GFAP阳性表达的细胞,FGFR3也呈阳性表达;术后1、3、5、7 d sham组大鼠脊髓FGFR3蛋白表达分别为(0.192 8±0.013 0)、(0.213 6±0.021 4)、(0.294 6±0.025 1)、(0.266 4±0.027 3),相应时间点SNI组大鼠脊髓FGFR3蛋白表达分别为(0.280 4±0.015 3)、(0.328 2±0.026 3)、(0.567 7±0.027 0)、(0.528 8±0.032 8),与sham组比较,SNI组脊髓FGFR3蛋白表达随时间增长而上调(P<0.05)。结论在神经病理性疼痛过程中FGFR3表达下调并可能参与GFAP的激活,提示FGFR3在神经病理性疼痛的发生以及发展过程中可能发挥着重要的作用。
Objective To observe the changes of the expression of fibroblast growth factor receptor 3 (FGFR3) in rat spinal cord after neurological pain in the selective sciatic nerve. Methods Forty SD male rats were divided into sham operation group and SNI group, with 20 rats in each group. The rat model of neuropathic pain was established by the selective ligation of the sciatic nerve branches. The sham group and the SNI group were observed on the 1st day before operation and on the 1st, 5th, 5th and 7th day after operation. The mechanical pain Thirty days after operation, the expression of FGFR3 and glial fibrillary acidic protein (GFAP) in spinal dorsal horn were observed by immunofluorescence double labeling. Results There was no significant difference in mechanical pain threshold between the two groups before operation (P> 0.05). The mechanical thresholds of rats in SNI group were (6.67 ± 2.12), (3.17 ± 0.99 ), (2.33 ± 0.65) and (1.75 ± 0.31) respectively. The mechanical thresholds of sham group were (12.75 ± 2.83), (12.33 ± 2.84), (12.08 ± 2.57) and (11.50 ± 2.65) The mechanical pain threshold of rats in SNI group was significantly lower than that of sham group at each time point (P <0.05). On the 7th day after surgery, the positive expression of GFAP in the dorsal horn of spinal cord was also observed in the SNI group. The expression of FGFR3 in the spinal cord of 7 d sham group were (0.192 8 ± 0.013 0), (0.213 6 ± 0.021 4), (0.294 6 ± 0.025 1) and (0.266 4 ± 0.027 3) respectively. The SNI group The expression of FGFR3 in the spinal cord of rats in SNI group was (0.280 4 ± 0.015 3), (0.328 2 ± 0.026 3), (0.567 7 ± 0.027 0) and (0.528 8 ± 0.032 8), respectively. Compared with sham group, The expression increased with time (P <0.05). Conclusions The expression of FGFR3 may be involved in the activation of GFAP during neuropathic pain, suggesting that FGFR3 may play an important role in the development of neuropathic pain.
