目的对吉林省2008年一起流行性腮腺炎(腮腺炎)爆发疫情进行病原学监测,观察是否出现典型的致细胞病变效应(Cytopathic Effect,CPE),并进行病毒核酸和基因型鉴定。方法采集病人的咽拭液,接种于生长良好的Vero/SLAM细胞(淋巴信号激活因子转染的非洲绿猴肾细胞),观察是否出现CPE,对三代病毒分离液采用逆转录-聚合酶链反应进行核酸鉴定,采用序列分析进行基因型鉴定。结果在采集到的15份标本中,有3份标本在Vero/SLAM细胞上可见到三种不同的典型CPE,6份标本核酸扩增为阳性,序列分析结果显示均为F基因型。结论在同一起爆发中,由同一株腮腺炎病毒引起的不同腮腺炎病例,虽然核苷酸序列完全一致,但所分离出的病毒在Vero/SLAM细胞上可表现为非常明显的不同类型的CPE。
Objective To etiologically monitor a mumps (mumps) outbreak in Jilin Province in 2008 to observe whether there is a typical cytopathic effect (CPE) and to identify the virus nucleic acid and genotype. Methods The pharyngeal swabs of patients were collected and inoculated into Vero / SLAM cells (African green monkey kidney cells transfected with lymphotactin) to observe whether there was CPE. Results Of the 15 samples collected, 3 samples showed typical CPE on Vero / SLAM cells, 6 samples showed positive amplification, and the results of sequence analysis showed that all of them were F genotypes. Conclusions In the same episode, different mumps cases caused by the same mumps virus, although the nucleotide sequences are identical, the isolated virus can show very different types of CPE on Vero / SLAM cells .
