目的:观察胃癌细胞内原癌基因erbB2表达降低前后信号转导与转录激活分子(STAT1)和细胞周期蛋白D1(cyclinD1)对表皮生长因子(EGF)刺激的反应性。方法:采用Westernbloting检测erbB2表达下调前后及EGF刺激前后STAT1和cyclinD1水平。结果:erbB2表达抑制对STAT1的EGF反应性无显著影响;erbB2表达下调后cyclinD1的EGF可诱导性显著降低。结论:erbB2表达降低可抑制EGF对cyclinD1的诱导效应。
Objective: To observe the response of STAT1 and cyclin D1 to epidermal growth factor (EGF) stimulation before and after the decrease of erbB 2 expression in gastric cancer cells. Methods: Western blotting was used to detect the expression of STAT1 and cyclinD1 before and after down-regulation of erbB2 expression and before and after EGF stimulation. RESULTS: The inhibition of erbB2 expression had no significant effect on the EGF reactivity of STAT1; the down-regulation of erbB2 expression significantly decreased EGF induction of cyclinD1. Conclusion: The decrease of erbB2 expression can inhibit the induction effect of EGF on cyclinD1.
