本文结合细胞培养和细胞遗传学方法研究急非淋在缓解非常早期(VER)血细胞中的残余白血病水平。作者选择4例奥尔小体阳性且带有(8:21)染色体易位的急非淋M_2型做研究。初诊时由静脉或骨髓收集原始细胞,DAT诱导化疗后在造血恢复时收集VER血细胞。同时做甲基纤维素和液体培养,培养体系中加入5—10%的植物血凝素刺激的白细胞条件培养液(PHA-LCM),培养3-14天收获细胞,部分涂片做形态学分析,部分进行染色体分析。结果:4例病人中只有2例可供VER期研究。初诊时采集的细胞在甲基纤维素和液体培养后均能检测到白血病相关的染色体易位与奥尔小体。所有的病例在培养3—9天后易检测到白血病核型。培养10天后,由于正常细胞超过白血病细胞,分化未期的细胞比例增加而奥尔小体阳性的原始细胞比例降低。在培养中不加PHA-LCM的细胞,只能检测到少量无t(8:21)的正常分裂中期。VER期血细胞在培养前后做形态学检查没有发现奥尔小体阳性的原始细胞。核型分析结果表明在大
This article combined cell culture and cytogenetic methods to investigate the effect of cisplatin on relieving residual leukemia in very early (VER) blood cells. The authors selected 4 cases of acute non-lymphocytic M 2 type with Orrish body positive and (8:21) chromosomal translocation. At the time of initial diagnosis, primitive cells were collected from veins or bone marrow, and VER blood cells were collected when hematopoiesis was restored after DAT induction chemotherapy. Simultaneously with methylcellulose and liquid culture, 5-10% phytohemagglutinin-stimulated leukocyte conditioned medium (PHA-LCM) was added to the culture system, cells were harvested for 3-14 days, and some smears were subjected to morphological analysis.
