以核桃楸外果皮为试材,采用超声辅助法及紫外可见分光光度法,研究了核桃楸外果皮总生物碱的最佳提取方法,并测定其抗氧化性和抑菌性,以期为核桃楸外果皮中生物碱的后续利用提供参考依据。结果表明:核桃楸外果皮生物碱最优提取条件为乙醇体积分数80%、料液比1∶10g·mL~(-1)、超声温度40℃、超声时间40min。抗氧化试验结果表明,核桃楸外果皮的总生物碱提取物对羟基自由基、亚硝酸根离子、超氧阴离子均有清除作用,由线性回归结果得到3种由基的IC_(50)值分别为0.030(·OH)、0.029(NO_2~-)、0.033g·L~(-1)(O_2~·)。抑菌试验结果表明,核桃楸外果皮的总生物碱提取物对木材褐腐菌密粘褶菌和白腐菌彩绒革盖菌的生长均存在抑制作用。
Using the outer skin of Juglans manshurica as test material, ultrasonic extraction and UV-Vis spectrophotometry were used to study the optimum extraction method of total alkaloids from Juglans mandshurica. The anti-oxidation and antimicrobial activities were determined. The exocarp alkaloids used in the follow-up to provide a reference. The results showed that the optimum extraction conditions were: ethanol volume fraction 80%, solid-liquid ratio 1:10 g · mL -1, ultrasonic temperature 40 ℃ and ultrasonic time 40 min. Antioxidant test results showed that total alkaloid extract of Juglans mandshurica skin had scavenging effects on hydroxyl radical, nitrite ion and superoxide anion. The IC 50 value of three kinds of base were obtained by linear regression respectively 0.030 (· OH), 0.029 (NO_2 ~ -) and 0.033g · L -1 (O_2 ~ ·), respectively. The results of antibacterial test showed that the total alkaloid extract of Juglans mandshurica skin had inhibitory effects on the growth of wood brown rot fungi and white rot fungi.
