目的综合评价解整合素金属蛋白酶33(A Disintegrin and Metalloproteinase 33,ADAM33)基因S2位点多态性与哮喘易感性的关系。方法制定统一的检索策略,检索Pubmed、Ovid-Medline、Embase、CNKI、维普及万方数据库中有关ADAM33基因多态性与哮喘易感性关系的病例-对照研究,按照纳入和排除标准选择文献提取相关信息,应用Review Manager 5.0软件进行Meta分析。结果纳入10篇合格文献,均满足遗传平衡检验,累计哮喘2 798例和对照4 357例,其中亚洲哮喘患者1 604例,欧美哮喘患者1 194例。Meta分析结果显示,ADAM33基因S2位点显性模型(CC+CG vs.GG)合并OR值为1.56(95%CI:1.01~2.40,P=0.04),按种族分层分析,亚洲哮喘人群OR值为2.15(95%CI:1.00~4.70,P=0.05)。结论 ADAM33基因S2位点多态性与亚洲人发生哮喘的易感性升高有一定相关性。
OBJECTIVE: To evaluate the relationship between allele S2 polymorphism of asthma and susceptibility to asthma in A Disintegrin and Metalloproteinase 33 (ADAM33) gene. Methods A unified search strategy was developed to search for case-control studies on the relationship between ADAM33 gene polymorphism and susceptibility to asthma in Pubmed, Ovid-Medline, Embase, CNKI, VIP and Wanfang databases, and to select relevant documents according to inclusion and exclusion criteria Information, Meta-analysis using Review Manager 5.0 software. Results Ten eligible articles were included in the study, all of which met the genetic balance test. There were 2 798 cases of asthma and 4 357 cases of control, including 1 604 cases of asthma in Asia and 1 194 cases of asthma in Europe and America. Meta-analysis showed that the combined odds ratio (OR) was 1.56 (95% CI: 1.01-2.40, P = 0.04) for ADAM33 gene S2 locus dominant model (CC + CG vs.GG) The value was 2.15 (95% CI: 1.00 to 4.70, P = 0.05). Conclusion There is a certain correlation between polymorphism of ADAM33 at S2 loci and increased susceptibility to asthma in Asians.
