目的:探讨化疗对骨肉瘤αV、β3整合素表达的影响。方法:免疫组化法检测20例化疗前和31例化疗后骨肉瘤αV、β3整合素的表达情况。结果:αV、β3整合素在骨肉瘤细胞呈强阳性表达,而正常骨细胞均呈阴性表达,P=0.000;化疗后两者表达明显下降,P=0.000。秩和检验表明,化疗后αV整合素与肿瘤Enneking分期(P=0.014)、侵袭(P=0.007)、复发(P=0.021)明显相关;β3整合素表达与肿瘤复发明显相关,P=0.003。多因素分析显示,化疗后αV整合素表达水平是影响骨肉瘤侵袭独立因素,化疗后β3整合素是影响骨肉瘤复发独立因素。结论:αV、β3整合素可能成为骨肉瘤诊断的辅助指标;化疗可能通过下调骨肉瘤细胞αV、β3整合素表达而改变其相应的生物学活性;αV、β3整合素可能分别是影响骨肉瘤侵袭、复发的独立因素;αV、β3整合素可能作为判断骨肉瘤化疗预后的指标及化疗药物作用的靶分子。
Objective: To investigate the effect of chemotherapy on the expression of αV and β3 integrins in osteosarcoma. Methods: Immunohistochemical method was used to detect the expression of αV, β3 integrin in 20 cases of osteosarcoma before chemotherapy and 31 cases of chemotherapy. Results: αV and β3 integrins were strongly expressed in osteosarcoma cells, while the normal osteoblasts were negative (P = 0.000). The expression of αV and β3 decreased significantly after chemotherapy (P = 0.000). The rank sum test showed that αV integrin was significantly correlated with Enneking stage (P = 0.014), invasion (P = 0.007) and recurrence (P = 0.021) after chemotherapy. The expression of β3 integrin was significantly correlated with tumor recurrence (P = 0.003). Multivariate analysis showed that the expression of αV integrin was an independent factor affecting the invasion of osteosarcoma after chemotherapy. Β3 integrin was an independent factor affecting the recurrence of osteosarcoma after chemotherapy. Conclusions: αV and β3 integrins may be auxiliary indicators for the diagnosis of osteosarcoma. Chemotherapy may alter the corresponding biological activity of αv and β3 integrins by down-regulating the expression of αV and β3 integrins in osteosarcoma cells. ΑV and β3 integrins may affect the invasion of osteosarcoma respectively , Recurrent independent factors; αV, β3 integrin may be used as a prognostic indicator of osteosarcoma chemotherapy and the role of target molecules.
