目的:研究天麻素对同型半胱氨酸诱导的小胶质细胞白介素-1β(IL-1β)、白介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)表达的影响,并对其结果与银杏叶提取物EGb761进行比较。方法:将体外培养的小鼠小胶质瘤细胞(BV-2细胞)分成空白对照组、同型半胱氨酸组、EGb761组(100mg/L)和天麻素低、中、高(2.5、5、10mmol/L)剂量组,培养72h。应用RT-qPCR方法评价IL-1β、IL-6和TNF-αmRNA表达,酶联免疫吸附法(ELISA)测定培养上清中上述细胞因子的蛋白浓度。结果:与对照组相比,同型半胱氨酸组细胞内IL-1β、IL-6和TNF-αmRNA表达和细胞培养上清液中蛋白浓度明显增加;与同型半胱氨酸组相比,EGb761组和天麻素各组细胞因子mRNA表达及培养上清液中蛋白浓度均降低,尤其是高剂量天麻素组与EGb761组作用相当,结果均具有统计学意义(P<0.05)。结论:天麻素能抑制同型半胱氨酸诱导的小胶质细胞IL-1β、IL-6和TNF-α表达,在一定浓度条件下其作用不亚于银杏叶提取物。
Objective: To investigate the effect of gastrodin on the expression of IL-1β, IL-6 and TNF-α in microglial cells induced by homocysteine The results were compared with EGb761, a Ginkgo biloba extract. Methods: The mouse microglial cells (BV-2 cells) cultured in vitro were divided into blank control group, homocysteine group, EGb761 group (100mg / L) and gastrodin low, medium and high , 10mmol / L) dose group, cultured for 72h. The expression of IL-1β, IL-6 and TNF-αmRNA was evaluated by RT-qPCR. The protein concentration of the above cytokines in the culture supernatant was determined by enzyme-linked immunosorbent assay (ELISA). Results: Compared with the control group, the protein levels of IL-1β, IL-6 and TNF-α in the cells of the homocysteine group and the cell culture supernatant were significantly increased. Compared with the homocysteine group, The mRNA expression of cytokines in EGb761 group and gastrodin group and the concentration of protein in culture supernatant decreased, especially in high-dose gastrodin group and EGb761 group. The results were statistically significant (P <0.05). Conclusion Gastrodin can inhibit the expression of IL-1β, IL-6 and TNF-α in microglial cells induced by homocysteine, which is as good as that of Ginkgo biloba extract at a certain concentration.
