目的研究放射线照射后NK-92细胞对人宫颈癌细胞的体外杀伤活性。方法应用医用电子直线加速器对NK-92细胞进行照射处理(0、400cGy)后,以体外培养人宫颈癌细胞系Hela为靶细胞,以NK-92的敏感细胞株K562作为对照,应用4h51Cr释放法检测不同效靶比情况下NK-92细胞的杀伤活性。结果NK-92细胞0cGy照射组,效靶比较低时(1∶1、5∶1)对Hela细胞杀伤率为15%~33%,随效靶比升高,杀伤率随之上升,10∶1时杀伤率显著上升为47%,20∶1时杀伤率仅上升了3%;对于K562细胞,杀伤率为33%~69%。400cGy组照射后2d,NK-92细胞对Hela细胞不同效靶比的杀伤率与0cGy组相比稍有降低,其总体杀伤率为14%~47%,对K562细胞杀伤范围在30%~60%之间。结论放射线照射后的NK-92细胞对人宫颈癌Hela细胞系具有一定的杀伤活性。
Objective To study the in vitro killing activity of NK-92 cells on human cervical carcinoma cells after radiation exposure. Methods NK-92 cells were treated with medical linear accelerator (0,400cGy) and cultured in vitro. Human cervical cancer cell line Hela was used as a target cell and NK-92-sensitive cell line K562 was used as a control. The 4h51Cr release method The killing activity of NK-92 cells was tested under different effective target ratios. Results The killing rate of Hela cells in NK-92 cells treated with 0cGy irradiation was 15% ~ 33% when the effective target was low (1: 1, 5: 1) 1 killing rate increased significantly to 47%, 20: 1 when the killing rate increased by only 3%; for K562 cells, the killing rate of 33% to 69%. The killing rate of NK-92 cells to Hela cells at 2 h after irradiation was slightly lower than that of 0 cGy group at 2 days after irradiation in the 400 cGy group, with an overall killing rate of 14% -47% and a K562 cell killing range of 30% -60 %between. Conclusion NK-92 cells after irradiation have certain cytotoxic activity on human cervical carcinoma Hela cell line.
