为探讨对纯中药制剂茅丹黄的抗弥漫性血管内凝血(DIC)作用和减轻脏器病理损害的机理,利用大肠杆菌内毒素 O_(127)B_6制兔 Shwartzman 反应模型。随机分为两组,治疗组以茅丹黄(2.0g/kg)灌胃4次。两组均于攻毒前后各取心血1次,分别检测血小板数、凝血酶原时间、部分凝血酶原时间、纤维蛋白原含量、血浆鱼精蛋白副凝试验、体外血栓、血清脂质过氧化物(LPO)浓度和血浆超氧化物歧化酶(SOD)活性等指标。结果:(1)茅丹黄具有明显抗 DIC 作用。(2)具有显著提高 SOD 活力和降低 LPO 水平的抗脂质过氧化物损伤作用(以上各项 P<0.05~0.001)。(3)抗内毒素血症。(4)明显减轻组织病理损害。本研究提供了茅丹黄活血化瘀、止血解毒和抗氧化自由基损伤等功效的另一理论依据。
In order to investigate the anti-diffuse intravascular coagulation (DIC) effect of pure Chinese medicinal preparation Maodanhuang and reduce the pathological mechanism of organ damage, a rabbit Shwartzman reaction model was made using the E. coli endotoxin O_(127)B_6. Randomly divided into two groups, the treatment group was fed with Mao Dan Huang (2.0g/kg) 4 times. Both groups took blood each time before and after the challenge to detect the number of platelets, prothrombin time, partial prothrombin time, fibrinogen content, plasma protamine paracoagulation test, in vitro thrombosis, serum lipid peroxidation LPO concentrations and plasma superoxide dismutase (SOD) activity and other indicators. Results: (1) Maodanhuang has obvious anti-DIC effect. (2) Anti-lipid peroxide injury with significant increase in SOD activity and decrease in LPO levels (P < 0.05-0.001 above). (3) Anti-endotoxemia. (4) Significantly reduce histopathological damage. This study provides another theoretical basis for the efficacy of Maodanhuang to activate blood circulation, stop bleeding, detoxify, and resist free radical damage.
