目的:建立千斤拔药材HPLC特征图谱分析方法。方法:采用高效液相色谱法,色谱柱为Waters Symmetry C18(4.6 mm×250 mm,5μm)柱,以乙腈(A)-水(B)为流动相,梯度洗脱(0~5 min,0%→5%A;5~10 min,5%→10%A;10~15 min,10%→30%A;15~25 min,30%→40%A;25~45 min,40%→80%A;45~80 min,80%A)。流速:0~5 min为0.6 mL·min-1,5~10 min为0.8 mL·min-1,10~80 min为1.0 mL·min-1。检测波长为254 nm,柱温为室温。以中药色谱特征图谱相似度评价系统的操作规范(2004 A版)软件计算。结果∶13批药材的相似度在0.506~0.926,确定了12个共有峰,建立了大叶千斤拔的HPLC特征图谱。结论:此方法简单、准确、重复性好,为千斤拔的质量控制提供了有效的方法。
Objective: To establish a method for the analysis of HPLC profiles of Qianjinba medicinal materials. METHODS: The HPLC was performed on a Waters Symmetry C18 (4.6 mm×250 mm, 5 μm) column with acetonitrile (A)-water (B) as the mobile phase. The gradient elution was performed (0 to 5 min, 0). %→5%A; 5~10 min, 5%→10%A; 10~15 min, 10%→30%A; 15~25 min, 30%→40%A; 25~45 min, 40%→ 80% A; 45-80 min, 80% A). The flow rate was 0.6 mL·min-1 for 0 to 5 min, 0.8 mL·min-1 for 5 to 10 min, and 1.0 mL·min-1 for 10 to 80 min. The detection wavelength is 254 nm and the column temperature is room temperature. The software was calculated with the operating specifications (2004 A version) of the chromatographic feature similarity evaluation system of traditional Chinese medicine. Results: The similarity of 13 batches of medicinal materials was between 0.506 and 0.926. Twelve common peaks were identified and the HPLC characteristic map of Daye Qianjin was established. Conclusion: This method is simple, accurate, and reproducible, providing an effective method for the quality control of the jacking horse.
