【目的】通过观察不同烫伤程度大鼠在烫伤24 h后β_1整合素和整合素连接激酶(ILK)的表达,探讨β_1整合素和ILK在创面愈合中的作用。【方法】将32只SD大鼠分别造成Ⅰ、浅Ⅱ、深Ⅱ度和Ⅲ度烫伤,伤后24 h取创面组织标本,制作切片,分别进行HE染色以及整合素β_1、ILK免疫组化染色,观察不同烫伤程度创面整合素β_1和ILK的表达与定位。【结果】Ⅰ度烫伤大鼠表皮基底层和皮肤附属器可见明显整合素β_1和ILK表达,其中ILK在皮肤附属器的表达较整合素β_1更为显著;浅Ⅱ度组织创面,残留的基底层和健全的皮肤附属器周围有整合素β_1和ILK表达,其中ILK在皮肤附属器的表达较整合素β_1更为显著,而整合素β_1主要在毛囊表达;深Ⅱ烫伤创面仅真皮深层健存的皮肤附属器中有整合素和ILK阳性细胞表达;Ⅲ度烫伤创面未见整合素β_1和ILK阳性。【结论】整合素β_1和ILK具有相似的局限表达模式并与大鼠烫伤程度密切相关,提示ILK可能介导整合素的胞内的信号通路,与整合素β_1共同参与创面愈合过程。
【Objective】 To investigate the effect of β 1 integrin and ILK on wound healing by observing the expression of β 1 integrin and integrin LK (ILK) in rats with different degrees of scald after 24 h scald. 【Methods】 Thirty-two SD rats were scalded for Ⅰ, Ⅱ, Ⅱ and Ⅲ degrees respectively. Twenty-four hours after injury, wound tissue samples were obtained and sliced. HE staining, integrin β 1 and ILK immunohistochemical staining The expression and localization of integrin β 1 and ILK in wounds with different degrees of scald were observed. 【Result】 The results showed that the expressions of integrin β 1 and ILK were obvious in the basal layer of the scalp and the skin appendages of scalded rats, and the expression of ILK in the skin adnexa was more significant than that of the integrin β 1. The superficial Ⅱ degree wounds, the residual basal layer And intact skin appendages have integrin β_1 and ILK expression. The expression of ILK in skin adnexa is more significant than that of integrin β_1, while that of integrin β_1 is mainly expressed in hair follicles. In deep Ⅱ scald wounds, the deep dermis There were integrin and ILK positive cells in skin appendages. No integrin β_1 and ILK were found in Ⅲ degree scald wounds. 【Conclusion】 Integrin β 1 and ILK have similar localized expression patterns and are closely related to the degree of scald in rats, suggesting that ILK might mediate the intracellular signaling pathways of integrin and participate in wound healing process with integrin β 1.
