应用AFLP技术对采自广东稻区7个地点不同生物型稻瘿蚊DNA进行指纹分析,并用DPS软件进行聚类分析,结果表明,AFLP可对稻瘿蚊DNA扩增出丰富的指纹带,不同稻瘿蚊种群和生物型DNA带型存在共同性,也有特异性;不同引物扩增数据用DPS软件分析得出不同的聚类结果。对4组引物的分析结果表明,只有1组引物所扩增的广东7个地点4个稻瘿蚊生物型DNA指纹可以进行较清晰聚类,说明鉴定稻瘿蚊生物型需要找到适合的引物组合,并指出利用DNA指纹技术分析稻瘿蚊生物型时存在一定的不确定性。
The AFLP technique was used to analyze the DNA of different biotypes of rice gall flies collected from 7 sites in the rice growing area of Guangdong Province. The cluster analysis was performed using DPS software. The results showed that AFLP could amplify DNA fingerprints of different species of rice gall midge, There is a common and specific genotypes between the Gallop mosquito population and the genotype DNA banding. The data of different primers were analyzed by DPS software to get different clustering results. The analysis of the 4 primer sets indicated that the DNA fingerprinting of 4 rice gall midge can be more clearly clustered in only one locus of Guangdong amplified by a group of primers, indicating that identification of the rice gall midge biotype requires finding a suitable primer combination , And pointed out that there is some uncertainty when using DNA fingerprinting to analyze the biology of rice gall midge.
