目的观察不同危险程度分组的急性淋巴细胞白血病(ALL)患儿血管内皮生长因子(VEGF)及其受体F lt-1和KDR的表达及与临床表现、疗效的关系。方法采用ELISA、RT-PCR方法检测正常儿童志愿者(20名)及ALL低危组(29例)、难治组(10例)、缓解组(20例)患儿VEGF及其受体表达,同时对不同分组ALL患儿的临床特点和指标进行参照对比分析。结果难治组患儿VEGF表达[(574.37±208.45)ng/L]高于低危组[(387.93±175.86)ng/L],后者又高于缓解组[(135.80±111.28)ng/L]和正常对照组[(91.16±41.34)ng/L],缓解组与正常对照组比较,差异无统计学意义;VEGF受体mRNA的RT-PCR检测结果从高至低依次为难治组、低危组、缓解组、正常对照组;不同分组VEGF及其受体表达结果与临床表现相吻合。结论高表达VEGF及其受体的患儿往往肿瘤负荷增高,尤其是在难治性ALL患儿中尤为明显,因此VEGF及其受体表达检测可能成为疾病预后的指标之一。
Objective To investigate the expression of vascular endothelial growth factor (VEGF) and its receptors F lt-1 and KDR in children with acute lymphoblastic leukemia (ALL) at different risk levels and their clinical manifestations and therapeutic effects. Methods The expressions of VEGF and its receptor were detected by ELISA and RT-PCR in normal children (20 children), ALL low-risk group (29 cases), refractory group (10 cases) and remission group (20 cases) At the same time, different groups of children with clinical features and indicators were compared with the comparative analysis. Results The expression of VEGF in the refractory group [(574.37 ± 208.45) ng / L] was significantly higher than that in the low-risk group [(387.93 ± 175.86) ng / L and 135.80 ± 111.28 ng / L ] And normal control group [(91.16 ± 41.34) ng / L]. There was no significant difference between the remission group and the normal control group. The RT-PCR results of VEGF receptor mRNA from high to low were refractory group, low Risk group, remission group and normal control group. The expression of VEGF and its receptor in different groups was consistent with clinical manifestations. Conclusion The high expression of VEGF and its receptor in children often have an increased tumor burden, especially in children with refractory ALL, especially the expression of VEGF and its receptor may be one of the prognostic indicators.
