目的 观察微囊化嗜铬细胞瘤细胞 (PC12细胞 )移植后帕金森病大鼠旋转行为变化。方法 采用免疫隔离微囊技术将能分泌多巴胺的 PC12细胞包裹后 ,移植到帕金森病大鼠脑内 ,观察阿扑吗啡诱发病鼠旋转行为变化 ,酪氨酸羟化酶(TH)抗体免疫组化染色观察移植区阳性细胞表达。结果 帕金森病大鼠旋转行为有明显改善。移植后 3月 ,大鼠脑内微囊完整 ,仍有 TH阳性 PC12细胞存活。结论 微囊化 PC12细胞移植可明显改善帕金森病大鼠的旋转行为。
Objective To observe the rotational behavior of Parkinson’s disease rats after transplantation of microencapsulated pheochromocytoma (PC12 cells). METHODS: Dopamine-secreting PC12 cells were encapsulated by immunoisolated microcapsules and transplanted into the brains of rats with Parkinson’s disease. The rotation behavior of apomorphine-induced rats was observed, and the tyrosine hydroxylase (TH) antibody immune group was observed. Staining was performed to observe the expression of positive cells in the transplanted area. Results The rotational behavior of Parkinson’s disease rats was significantly improved. Three months after transplantation, rat brain microcapsules were intact, and TH positive PC12 cells were still alive. Conclusion Transplantation of microencapsulated PC12 cells can significantly improve the rotational behavior of Parkinson’s disease rats.
