目的建立超高效液相色谱法测定钩吻素子的含量,并考察其水溶液在室温条件下的稳定性。方法采用Agilent Extend-C18(2.1 mm×100 mm,1.8μm)色谱柱,以乙腈-0.2%正丁胺水溶液(30∶70)为流动相,流速0.5 m L·min?1,检测波长256 nm,测定3个批次钩吻素子的含量,并考察钩吻素子水溶液在室温条件下稳定性。结果钩吻素子分离度良好;在10.19~112.2μg·m L?1内线性关系良好(r=0.999 7),定量限为0.1μg·m L?1;平均回收率为100.4%(RSD=0.45%,n=9);3个批次钩吻素子平均含量为99.97%;钩吻素子水溶液室温条件下连续7 d稳定性良好。结论该方法简单易行、专属性强,准确可靠,适用于钩吻素子含量测定,且本品水溶液稳定性良好。
OBJECTIVE To establish a HPLC method for the determination of koumissin and investigate the stability of aqueous solution at room temperature. Methods The mobile phase was acetonitrile-0.2% n-butylamine (30:70) with an Agilent Extend-C18 (2.1 mm × 100 mm, 1.8 μm) column at a flow rate of 0.5 m L · min -1. The detection wavelength was set at 256 nm , The determination of three batches of kiss kinin content, and check the kiss kiss kinsin aqueous solution at room temperature stability. The results showed that the kinki had a good resolution. The linear range was 10.19 ~ 112.2μg · m L -1 (r = 0.999 7) and the limit of quantification was 0.1μg · m L -1. The average recovery was 100.4% (RSD 0.45 %, n = 9). The average content of kouinkin in three batches was 99.97%. The aqueous solution of kougenin was stable for 7 days at room temperature. Conclusion The method is simple, specific, accurate and reliable. It is suitable for the determination of kojinkin, and the stability of the aqueous solution is good.
