目的探讨基于IS6110的PCR方法在唐山市结核分枝杆菌基因分型中的应用价值,分析唐山市结核分枝杆菌分离株的基因型分布特征。方法 2012年1月—2013年3月从唐山市第四医院就诊的结核病患者中分离结核分枝杆菌166株,采用基于IS6110的PCR方法进行基因分型。结果唐山市结核分枝杆菌基因型以北京基因型为主导。166株结核分枝杆菌具有明显的多态性,Ⅰ型、Ⅱ型、Ⅲ型分别包含51株、88株、11株,其他型别呈散在分布。在优势基因型(Ⅱ型)中,30~60岁组高于其他年龄组;农村患者比例偏高。结论基于IS6110的PCR方法能有效进行唐山市结核分枝杆菌的基因分型,证实唐山市结核分枝杆菌呈聚集性分布。
Objective To investigate the value of IS6110-based PCR in the genotyping of Mycobacterium tuberculosis in Tangshan and to analyze the genotype distribution of Mycobacterium tuberculosis isolates in Tangshan. Methods From January 2012 to March 2013, 166 strains of Mycobacterium tuberculosis were isolated from patients with TB in the Fourth Hospital of Tangshan City, and genotyped by PCR method based on IS6110. Results The genotypes of Mycobacterium tuberculosis in Tangshan were mainly Beijing genotypes. 166 strains of Mycobacterium tuberculosis have obvious polymorphism, type Ⅰ, type Ⅱ, type Ⅲ contain 51 strains, 88 strains, 11 strains, other types were scattered. In the predominant genotype (type II), the 30-60 age group was higher than the other age groups; the proportion of rural patients was high. Conclusion IS6110-based PCR method can effectively carry out the genotyping of Mycobacterium tuberculosis in Tangshan, confirming the aggregation distribution of Mycobacterium tuberculosis in Tangshan.
