背景:近期相关研究发现精氨酸酶Ⅰ与多种心血管疾病发生发展有关, 但国内外相关研究大都集中在动脉, 并未对静脉性疾病, 进行广泛研究。目的:观察深静脉血栓形成模型大鼠精氨酸酶Ⅰ的表达变化, 分析其在深静脉血栓形成过程中可能发挥的作用。方法:将100只SD大鼠随机分为正常对照组和模型组,采用血管钳夹股静脉+双后肢髋人字石膏外固定制动的方式建立大鼠创伤性深静脉血栓模型, 根据不同的观察时间点和血栓形成的不同病理生理过程情况分为血栓形成前组、血栓形成高峰期组和无血栓形成组,提取各组血液RNA,应用实时荧光定量 PCR 检测精氨酸酶Ⅰ在各组血细胞中的表达变化。结果与结论:血栓形成高峰期组精氨酸酶Ⅰ表达量比其他3组明显上升(P <0.01),正常对照组、血栓形成前组和无血栓形成组精氨酸酶Ⅰ表达差异无显著性意义 (P>0.05)。提示精氨酸酶Ⅰ与深静脉血栓形成密切相关。
BACKGROUND: Recent studies have found that arginase Ⅰ is involved in the development of a variety of cardiovascular diseases. However, most studies at home and abroad focus on arteries and do not study venous diseases extensively. Objective: To observe the changes of arginase Ⅰ in rat model of deep venous thrombosis and to analyze its possible role in the process of deep vein thrombosis. Methods: A total of 100 SD rats were randomly divided into normal control group and model group. Traumatic deep vein thrombosis model was established by means of vascular prosthesis femoral vein plus double hind limb hip implants immobilization. According to different The observation of time points and different pathophysiological process of thrombosis were divided into pre thrombosis group, peak thrombosis group and no thrombosis group, blood RNA was extracted from each group, real-time fluorescent quantitative PCR was used to detect arginase Ⅰ in each group Changes in the expression of blood cells. RESULTS AND CONCLUSION: Arginase Ⅰ expression in the peak thrombosis group was significantly higher than the other three groups (P <0.01). There was no significant difference in arginase Ⅰ expression between the normal control group, pre-thrombosis group and non-thrombosis group Sexual significance (P> 0.05). Tip arginase Ⅰ and deep vein thrombosis are closely related.
