目的 研究野生型PTEN对胶质母细胞瘤基因表达的影响及意义。方法 用表达野生型PTEN的载体转染PTEN失活的胶质母细胞瘤细胞系U87MG ,筛选出稳定表达野生型PTEN的细胞克隆 ,然后用cDNA微阵列技术找出野生型PTEN转染后mRNA表达改变的基因。结果 转染的野生型PTEN可以明显抑制U87MG肿瘤细胞的增殖。用cDNA微阵列技术分析后 ,共有 89个cDNA克隆在PTEN转染细胞与对照组细胞之间差异有显著性 (P <0 .0 1)。其中 ,76个为已知基因 ,包括胶质纤维酸性蛋白、p2 1/WAF1、转化生长因子β诱导的早期蛋白、DNA碎裂因子 4 5等 ;13个为未知基因。 结论 野生型PTEN可以影响多种基因的表达 ,从而调控胶质母细胞瘤细胞的增殖、分化、凋亡等过程
Objective To study the effect of wild type PTEN on the gene expression of glioblastoma and its significance. Methods PTEN-inactivated glioblastoma cell line U87MG was transfected with wild-type PTEN vector, and the cell clone stably expressing wild-type PTEN was screened. Then, the mRNA expression of wild-type PTEN after transfection was detected by cDNA microarray Changed genes. Results Transfection of wild-type PTEN significantly inhibited the proliferation of U87MG tumor cells. After analysis by cDNA microarray, a total of 89 cDNA clones had significant difference between PTEN transfected cells and control cells (P <0.01). Among them, 76 were known genes, including glial fibrillary acidic protein, p2 1 / WAF1, transforming growth factor β-induced early protein, DNA fragmentation factor 45 and so on; 13 were unknown genes. Conclusion Wild-type PTEN can affect the expression of multiple genes and thus regulate the proliferation, differentiation and apoptosis of glioblastoma cells
