目的 比较 3种不同的抗肿瘤药物 (喜树碱、胺苯吖啶、长春花碱 )对培养的淋巴细胞性 (Molt 4)或髓样白血病细胞 (HL 60 )的细胞周期的影响。方法 在体外用喜树碱、胺苯吖啶或长春花碱处理诱导HL 60或Molt 4细胞 3、6、9h后收获 ,75 %的冷乙醇固定 2 4h以上。用溴化丙啶对细胞核DNA染色 ,FACSort流式细胞仪分析细胞周期并依据Sub G1法计算G1、S、G2 /M期细胞或凋亡细胞的百分率。结果 喜树碱能选择性地诱导HL 60细胞的S和G2 /M期细胞凋亡 ,总凋亡率可达 41.2 7% ;胺苯吖啶仅能使部分Molt 4细胞阻滞于G2 /M期 ,阻滞率为 9.2 4% ;长春花碱使Molt 4细胞阻滞于G2 /M期阻滞率为 11.3 0 %。结论 HL 60细胞对喜树碱的细胞毒作用异常的敏感 ,易于发生凋亡 ;胺苯吖啶诱导Molt 4细胞阻滞于G2 /M期 ;长春花碱诱导Molt 4细胞阻滞于有丝分裂前期。
Objective To compare the effects of three different antitumor drugs (camptothecin, acamprosate, vinblastine) on the cell cycle of cultured lymphoid cells (Molt 4) or myeloid leukemia cells (HL 60). Methods HL 60 or Molt 4 cells were harvested after being treated with camptothecin, acridine or vinblastine for 3, 6 and 9 hours in vitro, and 75% cold ethanol was fixed for more than 24 hours. The nuclear DNA was stained with propidium bromide, the cell cycle was analyzed by FACSort flow cytometry, and the percentage of G1, S, G2 / M phase or apoptotic cells was calculated by Sub G1 method. Results Camptothecin selectively induced apoptosis of HL-60 cells at S and G2 / M phase, with a total apoptotic rate of 41.2 7%. Aminobenzidine blocked only part of Molt 4 cells at G2 / M The blocking rate was 9.24%. Vinblastine blocked the Molt4 cells in G2 / M phase with the blocking rate of 11.3%. CONCLUSION: HL-60 cells are abnormally sensitive to camptothecin. The apoptosis of Molt 4 cells induced by acamprosine in G2 / M phase is inhibited by amikacin. Vinblastine induces the arrest of Molt 4 cells in pre-mitosis.
