[目的]研究正常黏膜、癌旁正常组织、胃癌上非甾体抗炎药物活化基因-1(NAG-1)的表达情况及胃癌的发生发展与NAG-1的关系。[方法]测定正常胃黏膜、癌旁组织正常黏膜及胃癌组织上的mRNA及NAG-1的表达情况,可分别采用RT-PCR、免疫组化SP法测定。[结果]NAG-1蛋白在正常胃黏膜组织、癌旁正常组织、胃癌组织上的表达水平均不同,其中在癌旁正常组织的表达水平最高,明显高于其他2种组织中NAG-1蛋白的表达水平(P<0.01)。其次为正常黏膜的NAG-1蛋白的表达水平;而胃癌组织中的NAG-1蛋白的表达水平是最低的。同时NAG-1mRNA的表达水平也具有相同的规律,NAG-1mRNA在癌旁组织中表达最高,在正常组织黏膜中次之,而在胃癌黏膜上的表达最低。NAG-1mRNA在分化水平不同的胃癌组织中的表达水平也是不同的,在低分化胃癌中的NAG-1表达要低于高分化胃癌组织(P<0.01)。且可发现,NAG-1蛋白质表达与肿瘤大小,胃癌的浸润程度、TNM分化无明显的关联(P>0.05)。[结论]胃癌的发生发展过程中NAG-1起着重要的作用,其正常水平的表达可能对癌细胞的增殖起着抑制的作用,而NAG-1表达减弱或丢失会使其抑癌作用减弱或者丧失。
[Objective] To investigate the expression of NAG-1 in normal mucosa, adjacent normal tissues and gastric cancer and the relationship between the occurrence and development of gastric cancer and NAG-1. [Method] The mRNA and NAG-1 expression of normal gastric mucosa, adjacent normal mucosa and gastric cancer tissues were determined by RT-PCR and immunohistochemical SP method respectively. [Results] The expression levels of NAG-1 protein in normal gastric mucosa tissues, adjacent normal tissues and gastric cancer tissues were all different. The expression level of NAG-1 protein in normal tissues was the highest, which was significantly higher than that in the other two tissues (P <0.01). Followed by the expression of NAG-1 protein in normal mucosa, while the expression of NAG-1 protein in gastric cancer tissues was the lowest. Meanwhile, the expression of NAG-1 mRNA also has the same pattern. The expression of NAG-1 mRNA is the highest in paracancerous tissue, followed by normal mucosa, and lowest in gastric mucosa. The expression level of NAG-1 mRNA in gastric cancer tissues with different differentiation levels is also different, and the expression of NAG-1 in poorly differentiated gastric cancer tissues is lower than that in well differentiated gastric cancer tissues (P <0.01). And we found that there was no significant correlation between NAG-1 protein expression and tumor size, gastric cancer infiltration and TNM differentiation (P> 0.05). [Conclusion] NAG-1 plays an important role in the carcinogenesis and progression of gastric cancer. The normal expression of NAG-1 may play an inhibitory role in the proliferation of cancer cells. However, the decrease or loss of NAG-1 may reduce its anti-tumor effect Or lost.
