背景:目前强直性脊柱炎的发病机制尚不清楚,其发生可能与遗传因素、环境因素、慢性感染和自身免疫功能紊乱等因素有关。目前普遍认为遗传学因素在发病机制中起着重要作用,而且强直性脊柱炎的发病具有明显的家族聚集以及民族间的差异现象,目前寻找不同地区、不同民族之间强直性脊柱炎的差异性基因成为研究热点之一。目的:应用基因表达谱芯片技术筛选出当地维吾尔族与汉族强直性脊柱炎患者差异性基因,并对这些差异性基因进行比较分析。方法:随机采集新疆维吾尔自治区中医医院风湿科处于活动期的维吾尔族、汉族强直性脊柱炎患者各5例,另外分别选取各3例健康志愿者为对照,提取外周血中的RNA,制备探针后用于表达谱芯片杂交,筛选强直性脊柱炎组织差异表达基因,并采用半定量RT-PCR法验证芯片结果。结果与结论:通过对维吾尔族强直性脊柱炎与汉族强直性脊柱炎患者差异性基因表达研究发现,两组之间筛选出差异有显著性意义的miRNA 20个(P<0.05);通过对维吾尔族强直性脊柱炎与汉族强直性脊柱炎患者靶基因与miRNA对应关系分析发现,相关的靶基因共有15个,这些靶基因相对应并存在一定的调控关系的miRNA有79个,这些基因主要为白细胞抗原类和白细胞介素类,均与人体免疫相关。结果证实,在中国新疆地区维吾尔族与汉族强直性脊柱炎患者之间,可筛选出与强直性脊柱炎相关多个差异表达基因。
BACKGROUND: At present, the pathogenesis of ankylosing spondylitis is not yet clear, and its occurrence may be related to genetic factors, environmental factors, chronic infection and autoimmune disorders. OBJECTIVE: To screen differentially expressed genes between Uygur and Han patients with ankylosing spondylitis by using gene expression microarray technique, and to compare and analyze these differentially expressed genes. Methods: Randomly collected 5 cases of Uygur and Han ankylosing spondylitis in rheumatology department of traditional Chinese medicine hospital of Xinjiang Uygur Autonomous Region, and 3 healthy volunteers were selected as controls respectively to extract RNA from peripheral blood to prepare probes After the expression of the chip for hybridization, screening of ankylosing spondylitis differentially expressed genes, and the use of semi-quantitative RT-PCR chip results. RESULTS AND CONCLUSION: According to the differential gene expression in Uygur ankylosing spondylitis and Han patients with ankylosing spondylitis, 20 differentially significant miRNAs were screened between the two groups (P <0.05) Family members of ankylosing spondylitis and Han patients with ankylosing spondylitis target gene and miRNA correspondence analysis found that there are a total of 15 related target genes, these target genes corresponding to and there are a certain degree of regulation of miRNA 79, these genes are Leukocyte antigens and interleukins are immune to the human body. The results confirmed that in Xinjiang Uygur and Han patients with ankylosing spondylitis, can be screened out with ankylosing spondylitis related to multiple differentially expressed genes.
