目的探讨神经管发育缺陷(NTD)发生过程中与正常组织差异表达的相关基因,并作初步功能分类。方法用含有1100余个已知基因的中密度芯片比较了胚胎9.5d、10.5d正常与同期NTD小鼠神经管组织的基因表达差异。结果通过比较正常E9.5d与E10.5d获得138个差异表达基因,E9.5d-NTD获得20个差异,E10.5d-NTD获得29个差异表达基因,根据基因功能分类包括细胞周期与凋亡相关基因,信号转导基因,转录与翻译调控,能量与代谢基因,热休克基因,基质与骨架蛋白等多种种类。结论实验证实多类基因参与了NTD的发生、发展过程,对相关基因群的进一步研究有助于阐释NTD的发病机制。
Objective To investigate the expression of genes associated with normal tissues during the development of neural tube defects (NTDs) and to make preliminary functional classification. Methods The gene expression of neural tube tissues of normal and NTD mice at embryonic day 9.5 and 10.5 days was compared with that of medium density chips containing more than 1,100 known genes. Results 138 differentially expressed genes were obtained by comparing normal E9.5d and E10.5d with 20 differences in E9.5d-NTD and 29 differentially expressed genes in E10.5d-NTD. According to gene function classification, including cell cycle and apoptosis Related genes, signal transduction genes, transcriptional and translational regulation, energy and metabolic genes, heat shock genes, matrix and scaffold proteins. Conclusion Experiments confirmed that many kinds of genes involved in the occurrence and development of NTD, further study of related gene groups help to explain the pathogenesis of NTD.
