在体外用不同浓度的Ni2 O3 多次处理人肺成纤维细胞 (HLF) ,并进行转化细胞的恶性鉴定———刀豆凝集素A(ConA)试验和软琼脂细胞培养试验。发现Ni2 O3 可诱导HLF细胞恶性转化 ,转化细胞生长速度加快 ,排列紊乱 ,失去接触抑制 ,呈交叉重叠生长 ,在实验浓度范围内 ,转化率呈剂量—反应关系。转化细胞可被低浓度ConA凝集 ,并在软琼脂内生长。提示Ni2 O3 有较强的诱导人肺成纤维细胞恶性转化的能力 ,具有致癌性
Human lung fibroblasts (HLFs) were treated with various concentrations of Ni2O3 in vitro and malignant transformation of the transformed cells was performed - the ConA assay and the soft agar cell culture assay. It was found that Ni2O3 could induce the malignant transformation of HLF cells, and the transformed cells grew faster, arranged disorderly, lost contact inhibition and showed overlapping growth. Within the experimental concentration range, the conversion rate showed a dose-response relationship. Transformed cells can be agglutinated with low concentrations of ConA and grown in soft agar. It is suggested that Ni2O3 has strong ability to induce malignant transformation of human lung fibroblasts and has carcinogenicity
