本文通过对致倦库蚊属抗有机磷机制的研究,发现致倦库蚊属的耐药性与其体内基因扩增的酯酶有关。 待测的致倦库蚊有PelRR,Pelss,CoL,TEMR,MRes等五株。将现场采集的CoL株在有机磷-酒精混合液中筛选培养,得到五代同源CoL系列。以λ-噬菌体为载体,构建CoL基因组DNA文库并进行放射自显影。通过示踪、提纯,显示10个estα_3阳性斑和6个estβ_1阳性斑,然后在聚丙烯酰胺凝胶上电泳。CoL,TEMR的estβ_1扩增片段不同,但都有一条电泳迁移速率相同的增宽β-萘基乙酸特异性酯酶带。
In this paper, the anti-organophosphorus mechanism of Culex quinquefasciatus was studied and found that the resistance of Culex quinquefasciatus was related to the esterase gene amplification in vivo. Culex pipiens pallets to be tested with PelRR, Pelss, CoL, TEMR, MRes and other five. CoL strains collected on site were screened and cultured in organophosphorus-alcohol mixture to obtain five generations of homologous CoL series. Using λ-phage as a vector, a CoL genomic DNA library was constructed and autoradiographed. By tracing, purification, 10 estα_3 positive plaques and 6 estβ_1 positive plaques were displayed and then electrophoresed on a polyacrylamide gel. CoL, TEMR estβ_1 amplified fragments are different, but have a the same electrophoretic mobility of broadening β-naphthylacetate specific esterase bands.
