目的探讨妊娠期高血压疾病早期筛查的血清标志物。方法选择2003-11-2004-12在第三军医大学附属大坪医院产前检查及住院分娩的产妇,采用表面加强激光解析电离飞行时间质谱(surface-enhancedlaserdesorption/ionizationtimeofflight-massspectrometry,SELDI-TOF-MS)技术比较妊娠期高血压疾病组(5例)和正常妊娠组(5例)在妊娠13~16周时血清蛋白质谱图的差异。结果在质荷比为1000~20000范围内,妊娠期高血压疾病组的血清蛋白质谱图中质荷比(M/Z)为1324、3942、13749的蛋白质的相对含量(1.61±0.10、32.63±6.64、1.08±0.18)明显高于正常妊娠组(1.31±0.22、14.89±3.24、0.56±0.12),差异有统计学意义(P<0.05);而质荷比为2824、3245的蛋白质的相对含量(9.70±2.40;0.52±0.35)明显低于正常妊娠组(13.40±2.75、1.36±0.42),差异有统计学意义(P<0.05)。结论血清蛋白质谱图存在差异的5个质荷比相对应蛋白质组成的蛋白质组有可能成为妊娠期高血压疾病早期筛查的血清标志物。
Objective To investigate serum markers of early screening of hypertensive disorder complicating pregnancy. Methods From November 2003 to December 2004, antenatal examination and hospital delivery were performed in Daping Hospital Affiliated to the Third Military Medical University. The surface-enhanced laser desorption / ionization time-dependent light-mass spectrometry (SELDI-TOF-MS) Technical comparison of serum protein profiles in gestational hypertension group (n = 5) and normal pregnancy group (n = 5) between the 13th and 16th weeks of gestation. Results The relative content of protein with mass-to-charge ratio (M / Z) of 1324, 3942, and 2749 in the serum protein profile of gestational hypertension group was 1.61 ± 0.10,32.63 ± 6.64,1.08 ± 0.18) was significantly higher than that of normal pregnancy group (1.31 ± 0.22, 14.89 ± 3.24,0.56 ± 0.12), the difference was statistically significant (P <0.05); while the relative content of protein with mass-to-charge ratio of 2824,3245 (9.70 ± 2.40; 0.52 ± 0.35) was significantly lower than that of the normal pregnancy group (13.40 ± 2.75,1.36 ± 0.42), the difference was statistically significant (P <0.05). Conclusion There are five proteomic groups with different mass-to-protein ratios in the serum protein profiles, which may be the serum markers for early screening of hypertensive disorder complicating pregnancy.
