目的:研究丙氨瑞林降低人卵巢癌CoC1/cDDP细胞线粒体膜电位(ΔΨm)与诱导其凋亡的关系。方法:用不同浓度的丙氨瑞林与CoC1/cDDP细胞一起培养48h,并设对照组。经瑞士-姬姆萨染色进行细胞凋亡形态学观察;流式细胞仪检测亚G1期细胞、AnnexinV+/PI-细胞的百分率及线粒体膜电位(ΔΨm)的变化;半定量RT-PCR法检测作用前后CoC1/cDDP细胞Caspase-3mRNA的表达。结果:CoC1/cDDP细胞经丙氨瑞林作用后亚G1期细胞及AnnexinV+/PI-细胞的百分率较对照组明显升高(P<0.01),并且出现明显的凋亡细胞形态的改变,而线粒体膜电位(ΔΨm)降低的细胞数百分率显著增加(P<0.01),两者间呈正相关关系(r=0.950,r=0.924,P<0.05);Caspase-3mRNA的表达较对照组明显增强(P<0.01),并且与亚G1期细胞、AnnexinV+/PI-细胞及ΔΨm降低的细胞数均呈正相关关系(r=0.909,r=0.897,r=0.909,P<0.05);并且上述变化均与丙氨瑞林的剂量有关(P<0.01)。结论:丙氨瑞林可通过降低人卵巢癌CoC1/cDDP细胞线粒体膜电位(ΔΨm),从而诱导其凋亡。
OBJECTIVE: To investigate the relationship between the reduction of mitochondrial membrane potential (ΔΨm) and the induction of apoptosis of alaprostine in human ovarian cancer CoC1 / cDDP cells. Methods: Different concentrations of ararelin and CoC1 / cDDP cells were cultured for 48h, and the control group. The morphology of apoptotic cells was observed by Swiss-Giemsa staining. The percentage of cells in sub-G1 phase, the percentage of Annexin V + / PI-cells and the mitochondrial membrane potential (ΔΨm) were detected by flow cytometry. CONCLUSION: Atrarelin may induce apoptosis by reducing the mitochondrial membrane potential (ΔΨm) of human ovarian cancer CoC1 / cDDP cells.
