目的:探讨雄黄中砷对大鼠脑组织能量代谢的影响。方法:将96只大鼠随机分成12组,每组8只,以0.5%CMC-Na水溶液为混悬介质,雄黄低、中、高剂量组分别按0.3、0.9、2.7 g/kg灌胃给予雄黄,对照组给予等体积0.5%CMC-Na,分别连续给药2、4、6 w。采用氢化物发生原子吸收法测定脑组织中砷含量,高效液相色谱法测定三磷酸腺苷二钠(ATP-2Na)含量。结果:随着雄黄剂量的增加和给药时间的延长,大鼠脑组织中砷含量逐渐增加,说明雄黄中的砷可通过血脑屏障在脑组织蓄积。给予雄黄2 w后,脑组织中ATP-2Na含量与对照组无明显差异;给予雄黄4 w后,脑组织中ATP-2Na含量与对照组比较显著增加;给予雄黄6 w后,脑组织中ATP-2Na含量与对照组比较显著降低。不同剂量组间无显著差异。结论:雄黄中砷可在脑组织蓄积,对大鼠脑组织能量代谢产生影响。
Objective: To investigate the effect of realgar on the energy metabolism of brain tissue in rats. Methods: Ninety-six rats were randomly divided into 12 groups with 8 rats in each group. 0.5% CMC-Na aqueous solution was used as the suspending medium. The low, middle and high doses of realgar were orally administered at 0.3, 0.9 and 2.7 g / kg respectively Realgar, the control group was given an equal volume of 0.5% CMC-Na, respectively, continuous administration of 2,4,6 w. The content of arsenic in brain tissue was determined by hydride generation atomic absorption spectrometry and the content of adenosine triphosphate (ATP-2Na) was determined by high performance liquid chromatography. Results: As the dose of realgar increased and the administration time prolonged, the content of arsenic in rat brain gradually increased, indicating that arsenic in realgar could accumulate in the brain through the blood-brain barrier. The content of ATP-2Na in brain tissue after given realgar for 2 w was not significantly different from that in control group. The content of ATP-2Na in brain tissue increased significantly after 4 w and the concentration of ATP -2Na content decreased significantly compared with the control group. No significant difference between different dose groups. Conclusion: Arsenic in realgar can accumulate in brain tissue and affect the energy metabolism of brain tissue in rats.
