目的探讨创伤性休克对兔肾组织不同时相中性粒细胞明胶酶相关脂质运载蛋白(NGAL)、肾损伤分子-1(Kim-1)与热休克蛋白70(HSP70)表达的影响。方法健康新西兰大白兔30只,随机分为对照组、0 h模型组、6 h模型组、12 h模型组和24 h模型组,每组6只。通过酶联免疫吸附法(ELISA)、蛋白质印迹法(Western-blot)与实时荧光定量聚合酶链反应(Real-time PCR)测定各组兔血液、尿液及肾组织中NGAL、Kim-1、HSP70的含量与mRNA的表达变化。结果与对照组比较,模型组各时相血液及尿液中NGAL、Kim-1及HSP70含量均升高,6 h开始增加,24 h时有所恢复,但仍为高表达,差异有统计学意义(P<0.05)。与对照组比较,模型组兔肾组织在不同时相NGAL、Kim-1及HSP70表达量均显著升高,在6 h时升高最明显,差异均有统计学意义(P<0.05)。结论 NGAL、Kim-1、HSP70与创伤性休克导致肾损伤的发生和发展相关。监测血液、尿液及肾组织中NGAL、Kim-1及HSP70的变化情况,有助于对创伤性休克导致肾损伤的判断和治疗。
Objective To investigate the effects of traumatic shock on the expression of NGAL, Kim-1 and HSP70 in neutrophils at different phases of renal allograft in rabbits. Methods Thirty healthy New Zealand white rabbits were randomly divided into control group, 0 h model group, 6 h model group, 12 h model group and 24 h model group, with 6 rats in each group. The levels of NGAL, Kim-1, IL-6 and TNF-α in blood, urine and kidney of each group were detected by ELISA, Western-blot and real- HSP70 content and mRNA expression changes. Results Compared with the control group, the contents of NGAL, Kim-1 and HSP70 in the blood and urine of each model group increased at 6 h, increased at 24 h, and recovered at 24 h, but the difference was statistically significant Significance (P <0.05). Compared with the control group, the expression of NGAL, Kim-1 and HSP70 in renal tissue of model group were significantly increased at different time phases, and the most significant increase at 6 h (P <0.05). Conclusion NGAL, Kim-1, HSP70 are associated with the occurrence and development of renal injury induced by traumatic shock. Monitoring the changes of NGAL, Kim-1 and HSP70 in the blood, urine and kidney tissue can help to judge and treat the renal damage caused by traumatic shock.
