目的:探讨加味温胆汤对D-半乳糖所致衰老大鼠脑自由基及超微结构的影响。方法:SD大鼠随机分为正常组、模型组(ScD-半乳糖48mg/kg/d)、加味温胆汤组(10g/kg.ig,Sc D-半乳糖48mg/kg/d),连续40天,取大脑皮质测定SOD、MDA、GSH-Px、MAO、LPF及透射电镜观察。结果:注射D-半乳糖后,脑组织SOD活性降低(P<0.01),MDA含量及MAO活性增高(P<0.05),而LPF及GSH-Px活性无明显变化(P<0.05)。加味温胆汤则可提高脑组织SOD及GSH-Px活性(P<0.05和P<0.05),降低MDA含量(P<0.05),抑制MAO活性(P<0.01)。超微结构观察表明,注射D-半乳糖后,脂褐素、溶酶体大小形态不规则,线粒体肿胀、变性、嵴断裂、空泡样变,核膜内褶断裂、核仁边移等衰老改变。加味温胆汤则可使上述变性程度减轻。结论:加味温胆汤可延缓D-半乳糖所致的大鼠衰老,抑制脂质过氧化物的形成,提高抗氧化酶的活性,这可能是该方抗脑衰老的作用机理之一。
Objective: To investigate the effect of Jiawei Wendan Decoction on the brain free radical and ultrastructure of aging rats induced by D-galactose. The ultrastructural observation showed that after injection of D-galactose, lipofuscin and lysosomes were irregular in size, mitochondria were swollen, degenerated, sacral breaks, vacuolar degeneration, nuclear membrane folds, nucleolar shifts, and other aging. change. Modified warm soup can reduce the above degree of degeneration. Conclusion: Modified Wendan Decoction can delay the senescence of rats induced by D-galactose, inhibit the formation of lipid peroxidation, and increase the activity of antioxidative enzymes. This may be one of the mechanisms of anti-brain aging.
