目的检测CCN1基因在结肠癌组织中的表达情况,探讨CCN1的表达与结肠癌患者临床病理特征的关系及意义。方法应用Real-time PCR及Western blot方法检测CCN1在4例结肠癌及相应癌旁正常组织中mRNA和蛋白表达水平的差异表达;采用免疫组化检测117例结肠癌石蜡组织中CCN1的表达情况,分析其与结肠癌临床病理特征的关系及预后意义。结果在4例结肠癌组织中CCN1基因在mRNA及蛋白水平的表达高于相应正常癌旁组织;免疫组化检测结果显示CCN1蛋白在结肠癌组织中的表达阳性率为87.2%(102/117),强阳性率为58.1%(68/117),CCN1蛋白在结肠癌组织中的表达水平高于相应癌旁组织;统计学分析结果显示,CCN1蛋白高表达与结肠癌临床分期、淋巴结转移及远处转移密切相关,而与性别、年龄、肿瘤部位、肿瘤大小及分化程度无显著相关性。结论CCN1在结肠癌组织中高表达,其表达水平与肿瘤进展密切相关,提示CCN1在肿瘤发生和转移中可能起到重要作用。
Objective To detect the expression of CCN1 in colon cancer tissues and to explore the relationship between the expression of CCN1 and clinicopathological features in patients with colon cancer. Methods Real-time PCR and Western blot were used to detect the differential expressions of CCN1 mRNA and protein in 4 cases of colorectal cancer and corresponding normal tissues. Immunohistochemistry was used to detect the expression of CCN1 in 117 cases of colon carcinomas. To analyze its relationship with clinical and pathological features of colon cancer and prognostic significance. Results The expression of CCN1 mRNA and protein in colorectal cancer tissues was higher than that in normal paracancer tissues. The immunohistochemical results showed that the positive rate of CCN1 protein in colon cancer tissues was 87.2% (102/117) , The strong positive rate was 58.1% (68/117). The expression of CCN1 protein in colon cancer tissues was higher than that in corresponding paracancerous tissues. The statistical analysis showed that the expression of CCN1 protein was correlated with the clinical stage, lymph node metastasis and distant Department of metastasis is closely related, but with gender, age, tumor location, tumor size and the degree of differentiation was not significantly correlated. Conclusion CCN1 is highly expressed in colon cancer tissues. The expression of CCN1 is closely related to tumor progression, suggesting that CCN1 may play an important role in tumorigenesis and metastasis.
