疣粒野生稻(O.granulata Nees et Arn ex watt)是分布于我国的3种最具研究和利用价值的野生稻资源之一.利用southern杂交和荧光原位杂交的方法,对水稻抗绿叶蝉基因Glh,抗褐飞虱基因Bph-3和抗白叶枯病基因xa-5在栽培稻(Oryza sativa L)和疣粒野生稻中的同源性和物理位置进行了比较分析.southern杂交结果表明,在疣粒野生稻中存在与水稻抗性基因连锁的RFLP标记的同源序列.RFLP标记筛选出来的3个BAC克隆怍专探针.在栽培稻和疣粒野生稻有丝分裂中期和减数分裂粗线期染色体中均检测到杂交信号.双色荧光原位杂交确定了3个BAC克隆中的2个(14E16和38J9)在疣粒野生稻同一对同源染色体的短臂上,且这2个克隆在栽培稻和疣粒稻染色体上还存在共线性.另一个BAC克隆(44B4)被定位到2个物种的另一染色体短臂末端.虽然栽培稻和疣粒野生稻的亲缘关系较远,在生物学特征和生态习性上有着极为明显的区别,但疣粒野生稻被检出的染色体在相对长度、臂比和BAC克隆在染色体上的相对位置与栽培稻的结果相似.
