采用MS、B_6、White、Norstog四种培养基及六种植物激素,配制成多种培养基,对授粉后20—25天的花生幼胚进行离体培养和再生研究。结果显示:MS、B_6为幼胚离体培养和发育较好的培养基。MS+BA1.0mg/L(单位下同)+NAA0.05+GA_20.02+CH600+PVP0.2%+蔗糖5%能有效促进幼胚发育成熟。小苗通过MS+BA2.0+PP_(333)0.2+CH600的壮苗培养后在1/2MS大量元素+MS微量元素及有机物+NAA2.0+BA0.1+CH400+蔗糖2%中培养,诱根率高达96.6%。小苗移栽14天内保持饱和湿度其成活率为80%。再生植株在自然条件下能开花、结实。
MS, B_6, White, Norstog four kinds of media and six plant hormones were used to prepare a variety of media. The in vitro culture and regeneration of peanut immature embryos 20-25 days after pollination were studied. The results showed that: MS, B_6 embryos immature embryos cultured and developed better medium. MS + BA1.0mg / L (the same below) + NAA0.05 + GA_20.02 + CH600 + PVP0.2% + sucrose 5% can effectively promote immature embryos to mature. The seedlings were cultivated by strong seedlings of MS + BA2.0 + PP_ (333) 0.2 + CH600 and cultured in 1 / 2MS macronutrient + MS trace elements and organic matter + NAA2.0 + BA0.1 + CH400 + sucrose 2% Rate up to 96.6%.
