[目的]比较不同肺灌洗液对百草枯(PQ)致兔肺损伤的治疗效果。[方法]30只家兔随机分为5组:百草枯染毒组、生理盐水灌洗组、氨溴索灌洗组、乌司他丁灌洗组及对照组,每组6只动物。前4组一次性灌胃50 mg/kg体重的百草枯,3个灌洗组在染毒24 h后,分别给予生理盐水、氨溴索、乌司他丁进行肺灌洗;对照组一次性灌胃等体积的生理盐水,24 h后再行生理盐水肺灌洗。采集各组动物染毒前、染毒后第1、3、7天血液进行血清学检测;染毒后第7天处死动物,取其右肺组织进行病理学观察。[结果]染毒后第3、7天各灌洗组血清髓过氧化物酶(MPO)、羟脯氨酸(HYP)、白介素-1(IL-1)、肿瘤坏死因子-α(TNF-α)均低于百草枯染毒组(均P<0.05)。染毒后第7天,氨溴索灌洗组血清MPO及IL-1含量低于生理盐水灌洗组(均P<0.05);氨溴索灌洗组HYP水平高于乌司他丁灌洗组(P<0.05),但低于生理盐水灌洗组(P<0.05);乌司他丁灌洗组TNF-α水平低于氨溴索灌洗组(P<0.05)。3个灌洗组肺组织病理改变较染毒组轻。[结论]肺灌洗疗法可降低家兔百草枯中毒所致急性肺损伤的炎症反应,乌司他丁灌洗及氨溴索灌洗效果优于单纯生理盐水灌洗。
[Objective] To compare the therapeutic effect of different pulmonary lavage fluid on lung injury induced by paraquat (PQ) in rabbits. [Methods] Thirty rabbits were randomly divided into five groups: paraquat-exposed group, saline lavage group, ambroxol group, ulinastatin lavage group and control group, with 6 animals in each group. The first 4 groups were given gavage once a day with 50 mg / kg body weight of paraquat, and the three groups were given saline, ambroxol and ulinastatin respectively for lung lavage 24 h after exposure. In the control group, Gavage the same volume of saline, 24 h after saline lavage. Blood samples of each group were collected for blood test on the 1st, 3rd, 7th day after being exposed to the virus. The animals were sacrificed on the 7th day and the right lung tissues were taken for pathological observation. [Results] The levels of serum MPO, HYP, IL-1 and TNF-α in each lavage group were significantly higher than those in control group α) were lower than that of paraquat poisoning group (all P <0.05). The levels of MPO and IL-1 in the ambroxol group were lower than those in the saline group on the 7th day (all P <0.05), and the levels of HYP in ambroxol group were higher than those of ulinastatin group (P <0.05), but lower than saline lavage group (P <0.05). The level of TNF-α in ulinastatin lavage group was lower than that of ambroxol group (P <0.05). The pathological changes of lung tissue in the three lavage groups were lighter than those in the exposed group. [Conclusion] Pulmonary lavage therapy can reduce the inflammatory reaction of acute lung injury caused by paraquat poisoning in rabbits. The effects of ulinastatin and ambroxol are superior to those of saline alone.
