目的建立茶叶中5种儿茶素检测方法,比较浙江产15种不同茶叶中5种儿茶素含量差异性。方法采用改进的高效液相色谱法测定了茶叶中5种儿茶素的含量,对浙江产15种不同茶叶进行了儿茶素含量检测。色谱柱为大连依利特BDS C18(250 mm×4.6 mm)柱;流动相为乙腈∶0.1%柠檬酸水溶液=10∶90;柱温30℃,波长278 nm。结果改进的HPLC方法测定5种儿茶素在10~320 mg·L-1范围内线性关系良好(r>0.999),平均回收率95%以上,精密度3%以下。浙江产15种不同茶叶中5种儿茶素含量变化差异性较显著。结论建议进一步研究和完善采摘、加工及贮存等流程,并制定出科学、合理的质量标准。
Objective To establish a method for the determination of 5 kinds of catechins in tea and compare the differences of 5 kinds of catechins in 15 kinds of tea produced in Zhejiang Province. Methods The content of 5 kinds of catechins in tea was determined by improved high performance liquid chromatography (HPLC), and the content of catechins in 15 kinds of tea in Zhejiang Province was determined. The column was Dalian ELITE BDS C18 (250 mm × 4.6 mm) column; the mobile phase was acetonitrile: 0.1% citric acid solution = 10:90; the column temperature was 30 ℃ and the wavelength was 278 nm. Results The HPLC method was used to determine the linearity of 5 kinds of catechins in the range of 10-320 mg · L-1 (r> 0.999). The average recoveries were over 95% and the precision was below 3%. There were significant differences in the contents of 5 catechins in 15 kinds of tea produced in Zhejiang. Conclusion It is suggested to further study and improve the processes of picking, processing and storage, and formulate scientific and reasonable quality standards.
