【目的】研究新生大鼠心室成纤维细胞条件培养液促进心肌细胞肥大的作用。【方法】成纤维细胞和心肌细胞培养。【结果】成纤维细胞条件培养液能明显增大心肌细胞的表面积 ,增加心肌细胞的蛋白含量和 [3 H] 亮氨酸 ([3 H] Leu)的掺入 ,上述作用以第 3天的条件培养液作用最强 ,具有剂量依赖性。内皮素A受体 (ETAR)拮抗剂BQ12 3 能部分阻断成纤维细胞条件培养液促进心肌细胞肥大作用 ,而血管紧张素Ⅱ (AngⅡ )的Ⅰ型受体拮抗剂CV11974和α 肾上腺素受体拮抗剂酚胺唑啉(regitin)却无此效果。百日咳毒素 (PTX)和蛋白激酶C(PKC)抑制剂staurosporine(ST)也能部分阻断成纤维细胞条件培养液促进心肌细胞肥大作用。【结论】成纤维细胞条件培养液 (FCGM)中含有促使心肌细胞肥大的物质 ,这些物质可能是内皮素 1(ET 1)等 ,培养液的促肥大作用可能与PTX敏感的G蛋白及PKC有关。
【Objective】 To investigate the effect of neonatal rat ventricular fibroblast conditioned medium on cardiomyocyte hypertrophy. 【Methods】 Fibroblasts and cardiomyocytes were cultured. 【Results】 Fibroblast conditioned medium significantly increased the surface area of myocardial cells, increased the protein content of myocardial cells and the incorporation of [3 H] leucine ([3 H] Leu) on day 3 Conditioned medium has the strongest effect in a dose-dependent manner. Endothelin A receptor (ETAR) antagonist BQ12 3 partially blocked the fibroblast conditioned medium to promote cardiomyocyte hypertrophy, whereas angiotensin Ⅱ type I receptor antagonist CV11974 and α-adrenergic receptor The antagonist regitin did not. Pertussis toxin (PTX) and protein kinase C (PKC) inhibitor staurosporine (ST) also partially blocked fibroblast conditioned medium to promote cardiomyocyte hypertrophy. 【Conclusion】 The fibroblast conditioned medium (FCGM) contains substances that promote the hypertrophy of cardiomyocytes. These substances may be endothelin 1 (ET 1). The promoting effect of culture medium may be related to PTX sensitive G protein and PKC .
