1969年以来,田边药厂已用固定化酶柱进行工业化连续生产L-氨基酸,固定化酶是由酶离子结合到DEAE-Sephadex上制得。该方法利用L-氨基酸和未水解的酰基-D-氨基酸洗脱液浓缩后的溶解度的不同而分离。因此,假如能够利用高浓度底物,可预料有减少要蒸发的溶液体积之优点。由于氨基酰化酶是通过离子结合吸附在DEAE-Sephadex上,因此若利用高浓度底物,就会释出一些酶。为了克服上述局限,以及制备价廉的固定化氨基酰化酶,该厂研究了各种固定化氨基酰化酶的方法,并报道了它的制备性质及适用性。最近发现,将丹宁共价结合到氨 Since 1969, Tanabe Pharmaceutical Factory has carried out industrial production of L-amino acids by immobilized enzyme columns, and immobilized enzymes are produced by binding of enzyme ions to DEAE-Sephadex.
