目的:检测喉鳞状细胞癌中磷酸化的信号转导和转录激活因子3(p-STAT3)表达与各临床病理指征的关系,并探讨与磷酸酶与张力蛋白同源物(PTEN)的相关性。方法:采用免疫组织化学法检测p-STAT3、PTEN蛋白在67例喉鳞状细胞癌及25例癌旁(>2.0cm)正常黏膜中的表达,探讨其相关性及与各临床病理指征的关系,并采用Kaplan-Merier法进行预后分析。结果:p-STAT3在喉鳞状细胞癌组织中的表达率为71.64%,在正常黏膜中的表达为16%,组间比较差异具有统计学意义(Zc=4.705 2,P<0.01);p-STAT3高表达与喉鳞状细胞癌的分化程度(Hc=14.928 6,P<0.01)、临床分期(Zc=12.437 5,P<0.01)、颈淋巴结转移(Zc=4.512 4,P<0.01)、预后(Zc=12.312,P<0.01)有关;PTEN在喉鳞状细胞癌组织和癌旁正常黏膜中阳性表达率分别为41.79%、96.00%,组间比较差异具有统计学意义(Zc=5.703 7,P<0.01);PTEN蛋白表达与喉鳞状细胞癌的临床分期(Zc=3.889 7,P<0.05)、分化程度(Hc=12.889,P<0.05)、颈部淋巴结转移(Zc=2.082 4,P<0.05)、预后(Zc=4.205 2,P<0.01)呈负相关。在喉鳞状细胞癌组织中p-STAT3和PTEN呈负相关关系,相关系数rs=-0.514 8(P<0.05),随访中5年存活患者p-STAT3阳性表达率56.25%(18/32)明显低于死亡患者82.35%(14/17)。结论:p-STAT3和PTEN蛋白表达与喉鳞状细胞癌的病情进展、转移及预后相关,p-STAT3和PTEN表达在喉鳞状细胞癌中呈负相关关系,PTEN可能是p-STAT3下游的靶基因。
Objective: To investigate the relationship between the expression of phosphorylated signal transducers and activators of transcription 3 (p-STAT3) and clinicopathological features in laryngeal squamous cell carcinoma (LSCC), and to explore the relationship between the expression of PTEN and phosphatase Correlation. Methods: The expressions of p-STAT3 and PTEN protein in 67 cases of laryngeal squamous cell carcinoma and 25 cases of para-carcinoma (> 2.0 cm) normal mucosa were detected by immunohistochemistry. The correlations between them and the clinicopathological parameters Relationship, and Kaplan-Merier method for prognosis analysis. Results: The positive rate of p-STAT3 in laryngeal squamous cell carcinoma was 71.64% and in normal mucosa was 16%, the difference was statistically significant (Zc = 4.705 2, P <0.01); p (Zc = 12.437 5, P <0.01), cervical lymph node metastasis (Zc = 4.512 4, P <0.01), and there was no difference between the expression of STAT3 and the degree of laryngeal squamous cell carcinoma (Hc = 14.928 6, (Zc = 12.312, P <0.01). The positive expression rate of PTEN in laryngeal squamous cell carcinoma and adjacent normal mucosa was 41.79% and 96.00%, respectively. The difference between the two groups was statistically significant (Zc = 5.703 7, P <0.01). The expression of PTEN protein was significantly correlated with the clinical stage of laryngeal squamous cell carcinoma (Zc = 3.889 7, P <0.05) 4, P <0.05), and prognosis (Zc = 4.205 2, P <0.01). There was a negative correlation between p-STAT3 and PTEN in laryngeal squamous cell carcinoma, the correlation coefficient rs = -0.5148 (P <0.05), and the positive expression rate of p-STAT3 in the 5-year follow-up was 56.25% (18/32) Significantly lower than the death of patients 82.35% (14/17). CONCLUSIONS: The expressions of p-STAT3 and PTEN are correlated with the progression, metastasis and prognosis of laryngeal squamous cell carcinoma. The expressions of p-STAT3 and PTEN are negatively correlated in laryngeal squamous cell carcinoma. PTEN may be downstream of p-STAT3 Target gene
